Overview

**S095029 + pembrolizumab** is a combination regimen under clinical investigation for the treatment of locally advanced, unresectable, or metastatic microsatellite instability-high (MSI-H) or defective mismatch repair (dMMR) gastric and gastroesophageal junction adenocarcinoma. S095029 (also known as SYM025) is an investigational monoclonal antibody targeting NKG2A, an inhibitory receptor expressed on natural killer (NK) cells and some T cells, aiming to enhance innate and adaptive anti-tumor immunity. Pembrolizumab is a marketed monoclonal antibody (KEYTRUDA) that blocks the PD-1 receptor, restoring immune recognition and attack of tumor cells. Patients in clinical trials receive both drugs via intravenous infusion every three weeks. The combination is being evaluated for safety, tolerability, and antitumor activity in Phase 1b/2 studies[1][2][3][4][5][6][7][8][10].
