S095035 + TNG462 is an investigational oral combination therapy for advanced or metastatic solid tumors with homozygous MTAP deletion. **S095035** is a methionine adenosyltransferase 2A (MAT2A) inhibitor, designed to disrupt the methionine cycle and reduce production of S-adenosylmethionine, a key metabolite in MTAP-deleted cancer cells. **TNG462** is a selective, MTA-cooperative PRMT5 inhibitor developed by Tango Therapeutics, which blocks protein arginine N-methyltransferase 5 (PRMT5) activity specifically in the presence of MTA, causing selective cytotoxicity in MTAP-deleted tumors with fewer effects on normal cells. The combination exploits vulnerabilities in the methionine cycle and PRMT5 methylation pathways present in MTAP-deleted cancers. Both agents are administered orally and are designed for patients who have exhausted standard therapies. The drug combo is currently being evaluated in a first-in-human, open-label, multicenter phase 1/2 trial across several solid tumor types, including non-small cell lung cancer, cholangiocarcinoma, pancreatic ductal adenocarcinoma, and gastroesophageal cancer[1][4][6][8][10].
