S65487 is an investigational small molecule that acts as a selective inhibitor of the Bcl2 protein, which plays a key role in regulating apoptosis (programmed cell death) in cancer cells. By inhibiting Bcl2, S65487 promotes apoptosis in malignant cells. Azacitidine is a pyrimidine nucleoside analogue and hypomethylating agent used for the treatment of myelodysplastic syndromes (MDS), acute myeloid leukemia (AML), and other hematologic malignancies. It works primarily by inhibiting DNA methyltransferase, leading to hypomethylation of DNA and reactivation of tumor suppressor genes, as well as direct cytotoxicity through incorporation into RNA and DNA. The combination of S65487 with azacitidine is being studied for its potential synergistic effects in treating adults with untreated AML who are not suitable for intensive chemotherapy[2][4][5][6].
