Overview

This is a combination regimen of **sabatolimab**, **magrolimab**, and **azacitidine**. - **Sabatolimab** is a monoclonal antibody targeting T-cell immunoglobulin and mucin-domain containing-3 (TIM-3), an immune checkpoint expressed on myeloid cells and leukemic stem cells. It is designed to enhance anti-tumor immunity by blocking TIM-3, thereby promoting immune-mediated clearance of malignant cells. - **Magrolimab** is a humanized monoclonal antibody against cluster of differentiation 47 (CD47), a "don't eat me" signal overexpressed on many cancer cells. By blocking CD47, magrolimab promotes macrophage-mediated phagocytosis of tumor cells. - **Azacitidine** is a nucleoside metabolic inhibitor and DNA methyltransferase inhibitor used to treat myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML). It works as a hypomethylating agent, reactivating tumor suppressor genes and promoting cytotoxicity against hypermethylated neoplastic cells. This triple combination is being investigated in clinical trials for **acute myeloid leukemia (AML)** and **higher-risk myelodysplastic syndromes (MDS)** to determine safety and preliminary efficacy in previously untreated or unfit patients for intensive chemotherapy[1][4][7].
