Overview

A combination of three investigational monoclonal antibodies—**sabatolimab** (MBG453), **NIS793**, and **spartalizumab** (PDR001)—developed by Novartis for cancer immunotherapy, primarily targeting myeloid and solid tumor diseases. Sabatolimab is a humanized anti-TIM-3 antibody that activates the immune system and targets leukemic stem cells, promising long-lasting responses in myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML)[1][3][5]. NIS793 is an anti-TGF-β monoclonal antibody under trial for solid tumors, aiming to block the immunosuppressive tumor microenvironment by antagonizing TGF-β signaling. Spartalizumab is a humanized IgG4 anti-PD-1 monoclonal antibody that blocks PD-1–mediated immune suppression, enhancing antitumor T-cell activity; it has shown efficacy and safety for multiple tumor types, including esophageal squamous cell carcinoma, nasopharyngeal cancer, and several solid tumors[2][4][6]. The combination is being investigated for synergistic checkpoint inhibition and immune activation in advanced malignancies.
