Sacituzumab govitecan + enfortumab vedotin is a combination of two antibody-drug conjugates (ADCs), sacituzumab govitecan and enfortumab vedotin, administered together as an investigational therapeutic regimen for metastatic urothelial carcinoma. Sacituzumab govitecan targets the cell surface antigen Trop-2 and delivers the cytotoxic payload SN-38, an active metabolite of irinotecan, inducing apoptosis in Trop-2–expressing tumor cells. Enfortumab vedotin targets Nectin-4 and delivers monomethyl auristatin E (MMAE), a microtubule-disrupting agent, which causes cell death in Nectin-4–expressing tumors. Both drugs leverage the specific targeting of tumor-associated antigens with potent cytotoxic activity for the treatment of advanced or treatment-resistant urothelial carcinoma. Phase I data indicate a favorable safety profile and promising objective response rates when used in combination, particularly in patients who have progressed following standard therapies[1][5][7].
