This combination therapy consists of sacituzumab govitecan, a Trop-2-directed antibody-drug conjugate (ADC), and fulvestrant, a selective estrogen receptor degrader (SERD). Sacituzumab govitecan is composed of a humanized monoclonal antibody targeting the trophoblast cell-surface antigen 2 (Trop-2) conjugated via a hydrolyzable linker to SN-38, the active metabolite of irinotecan and a potent topoisomerase I inhibitor. Fulvestrant is a small molecule that binds to and degrades the estrogen receptor, thereby inhibiting hormone-driven tumor proliferation. This specific combination is being evaluated by the Hunan Cancer Hospital in the Phase Ib/II SoGreat trial for patients with hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) metastatic breast cancer who have previously progressed on CDK4/6 inhibitors.
