sacituzumab govitecan + nivolumab + relatlimab is an experimental combination therapy being investigated for the treatment of metastatic triple-negative breast cancer (mTNBC). This regimen combines **sacituzumab govitecan**, a Trop-2-directed antibody-drug conjugate (ADC) that delivers a cytotoxic topoisomerase I inhibitor payload (SN-38), with dual immune checkpoint inhibition provided by **nivolumab** (targeting PD-1) and **relatlimab** (targeting LAG-3). The therapeutic strategy aims to leverage the ADC's ability to induce immunogenic cell death and reduce tumor burden while simultaneously overcoming T-cell exhaustion and immune evasion through the inhibition of the PD-1 and LAG-3 pathways. This triple combination is currently being evaluated in the Phase Ib SIMONE trial, led by Yale University, as a second-line therapy for patients with mTNBC.
