Drug intelligence / Profile preview
Sacituzumab govitecan + trastuzumab is an investigational combination regimen consisting of the TROP2-directed antibody-drug conjugate (ADC) sacituzumab govitecan and the HER2-targeted monoclonal antibody trastuzumab. Sacituzumab govitecan delivers the topoisomerase I inhibitor payload SN-38 to TROP2-expressing tumor cells, while trastuzumab binds to HER2 to inhibit tumor cell proliferation and mediate antibody-dependent cellular cytotoxicity (ADCC). The combination has been evaluated in the phase II SATEEN trial (NCT06100874) for the treatment of patients with heavily pretreated HER2-positive metastatic breast cancer who progressed on trastuzumab deruxtecan (T-DXd). However, the trial failed to meet its primary endpoint of objective response rate (ORR), demonstrating limited clinical activity and significant hematologic toxicity.
