Drug intelligence / Profile preview
Sacituzumab tirumotecan + pembrolizumab + bevacizumab is an investigational combination regimen being evaluated for the treatment of advanced cancers, specifically as a first-line maintenance therapy for persistent, recurrent, or newly diagnosed metastatic cervical cancer with PD-L1 combined positive score (CPS) ≥1. The regimen combines sacituzumab tirumotecan (sac-TMT; MK-2870; SKB264), a trophoblast glycoprotein (TROP2)-directed antibody-drug conjugate (ADC) carrying a topoisomerase I inhibitor payload; pembrolizumab (Keytruda), a programmed cell death protein 1 (PD-1)-blocking monoclonal antibody; and bevacizumab (Avastin), a vascular endothelial growth factor (VEGF)-targeting monoclonal antibody. This combination is designed to simultaneously target tumor cells via TROP2, enhance anti-tumor immune responses by blocking the PD-1/PD-L1 pathway, and inhibit angiogenesis by targeting VEGF. It is currently being evaluated in the Phase 3 TroFuse-036/GOG-3123/ENGOT-cx22 clinical trial.
