Drug intelligence / Profile preview
This is a combination therapy consisting of four drugs: - **Sacituzumab tirumotecan** is an antibody-drug conjugate (ADC) targeting TROP2. It delivers a cytotoxic topoisomerase I inhibitor payload to TROP2-expressing cancer cells, leading to cell death[3][4][5]. - **Pembrolizumab** is a humanized monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1), thereby enhancing the immune response against tumor cells. It is classified as an immune checkpoint inhibitor and is approved for multiple cancers[4][6][7]. - **Carboplatin** is a platinum-based chemotherapeutic agent that crosslinks DNA, preventing cell division and inducing apoptosis in cancer cells. - **Paclitaxel** is a microtubule inhibitor that stabilizes microtubules and prevents their disassembly, thereby inhibiting mitosis and promoting apoptosis in cancer cells. This particular four-drug combination is under investigation for the treatment of triple-negative breast cancer (TNBC), especially in the neoadjuvant (pre-surgical) or adjuvant (post-surgical) settings and is being evaluated in phase 2/3 clinical trials[3][4][6]. Sacituzumab tirumotecan is developed by Merck (MSD), with SKB264 as a name used in some trials; pembrolizumab is developed and manufactured by Merck (MSD); carboplatin and paclitaxel are established chemotherapeutic agents produced by numerous generic manufacturers.
