Overview

sacituzumab tirumotecan + toripalimab is a combination therapy being investigated for the treatment of non-small cell lung cancer (NSCLC). It consists of sacituzumab tirumotecan (also known as SKB264 or MK-2870), a trophoblast cell-surface antigen 2 (TROP2)-directed antibody-drug conjugate (ADC), and toripalimab, a monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. Sacituzumab tirumotecan utilizes a belotecan-derivative topoisomerase I inhibitor payload conjugated via a novel linker to a TROP2-targeting antibody, designed to deliver cytotoxic effects directly to tumor cells. Toripalimab acts as an immune checkpoint inhibitor, enhancing the body's immune response against cancer cells. This specific combination is being evaluated in a Phase II clinical trial sponsored by Hunan Province Tumor Hospital as an induction and consolidation therapy for patients with initially unresectable stage III NSCLC.
