SA-TPP + C10 is a combination of two mitochondrial-targeted lipophilic cations: salicylate-triphenylphosphonium (SA-TPP) and decyl-triphenylphosphonium (C10-TPP). These compounds utilize the triphenylphosphonium (TPP+) moiety to selectively accumulate within the mitochondrial matrix of cells with high mitochondrial membrane potential, such as cancer cells and tumor-initiating cells (TICs). SA-TPP is a mitochondrial-targeted derivative of salicylate that uncouples oxidative phosphorylation and disrupts mitochondrial metabolism. C10-TPP is a decyl-substituted TPP cation that serves to further destabilize mitochondrial membranes and organelle function. This combination is being investigated as a therapeutic strategy to overcome drug resistance and eradicate cancer stem cells in hypoxic non-small cell lung carcinoma (NSCLC). It is frequently studied in synergy with the mitoribosome inhibitor doxycycline to induce a metabolic catastrophe by simultaneously inhibiting mitochondrial function and biogenesis.
