Overview

Samuraciclib + anti-androgen is a combination therapy in development by Carrick Therapeutics for the treatment of castration-resistant prostate cancer (CRPC). The regimen consists of samuraciclib (CT7001), an orally bioavailable, small molecule ATP-competitive inhibitor of cyclin-dependent kinase 7 (CDK7), and an anti-androgen agent (such as enzalutamide). CDK7 is a key regulator of the cell cycle and global transcription, and it has been shown to specifically promote androgen receptor (AR) signaling. By simultaneously inhibiting CDK7 and the AR pathway, the combination aims to suppress AR-mediated transcription and overcome resistance mechanisms in prostate cancer cells. Preclinical studies have demonstrated that this combination significantly augments growth inhibition and induces apoptosis compared to monotherapy. The combination is currently being evaluated in Phase 1 clinical trials for prostate cancer.
