Sapanisertib + everolimus is a **combination regimen** of two oral small molecule inhibitors targeting the mTOR (mechanistic target of rapamycin) pathway. **Sapanisertib** is a dual mTORC1/mTORC2 inhibitor, designed to more completely inhibit the PI3K/AKT/mTOR oncogenic signaling cascade than first-generation mTOR inhibitors, which only inhibit mTORC1. **Everolimus** is a selective mTORC1 inhibitor, approved for various cancers including renal cell carcinoma and breast cancer. This combination has been explored in clinical trials for enhanced pathway suppression to overcome resistance mechanisms in advanced or metastatic cancers, particularly clear cell renal cell carcinoma.
