sapitinib + paclitaxel

AZD8931 + paclitaxel is an investigational combination therapy developed by AstraZeneca, pairing the pan-ErbB inhibitor sapitinib (AZD8931) with the microtubule-stabilizing chemotherapy agent paclitaxel. Sapitinib is an oral, reversible, and equipotent small molecule inhibitor of the epidermal growth factor receptor (EGFR), HER2, and HER3 signaling pathways. The combination was primarily evaluated for the treatment of advanced solid tumors and breast cancer, particularly in patients with low HER2 expression. In the Phase II THYME trial, the addition of sapitinib to weekly paclitaxel failed to significantly prolong progression-free survival in women with low HER2-expressing advanced breast cancer, despite showing an increase in objective response rate.
