SAR444881 + cetuximab is a combination therapy of SAR444881, a novel humanized IgG4 monoclonal antibody targeting ILT2 (Leukocyte immunoglobulin-like receptor B1), and cetuximab, an epidermal growth factor receptor (EGFR) inhibitor monoclonal antibody. SAR444881 selectively blocks ILT2, which interacts with HLA-G on tumor cells to suppress immune function. By inhibiting ILT2, SAR444881 disrupts the "don't eat me" signal on tumor cells, restoring and enhancing anti-tumor activities of macrophages, NK cells, and T cells through increased phagocytosis, cytotoxicity, and cytokine secretion. Cetuximab specifically blocks EGFR to inhibit tumor cell growth and proliferation. The combination aims to synergistically promote immune-mediated tumor clearance and inhibit tumor growth. SAR444881 was initially developed by Biond Biologics and further developed and sponsored by Sanofi. This regimen is under investigation for advanced solid tumors, including colorectal cancer (CRC), non-small cell lung cancer (NSCLC), gastric cancers, and other malignancies[1][3][4][5][7][9].
