Overview

Sargramostim + ipilimumab is a combination therapy used primarily in advanced melanoma. Sargramostim is a recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF). It enhances antigen presentation and boosts antitumor activity through stimulation of dendritic cells and T/B lymphocytes. Ipilimumab is a fully human IgG1 monoclonal antibody targeting cytotoxic T lymphocyte antigen-4 (CTLA-4), increasing immune-mediated attack against tumor cells. In clinical studies, this combination improved overall survival and reduced high-grade immune-related adverse events compared to ipilimumab alone. Its mechanism integrates immune checkpoint blockade with cytokine-mediated enhancement of immune response[1][2][3][4][5].
