A three-drug investigational oncology regimen combining sargramostim, thalidomide, and docetaxel. Sargramostim is recombinant human granulocyte-macrophage colony-stimulating factor that activates and matures dendritic cells and enhances antitumor immune responses. Thalidomide is an immunomodulatory and antiangiogenic small molecule that inhibits pro-angiogenic signaling and modulates T cell and NK cell function. Docetaxel is a taxane chemotherapeutic that stabilizes microtubules to inhibit mitosis and induce apoptosis, with additional antiangiogenic effects reported. The combination has been explored primarily in prostate cancer to augment chemotherapy with immunostimulation and antiangiogenesis; early-phase work studied sargramostim plus thalidomide and separately thalidomide plus docetaxel, with DrugBank listing a terminated phase 2 trial including all three components in prostate cancer.
