Sargramostim + trastuzumab is an investigational combination therapy consisting of two biologic agents. Sargramostim is a recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF) that stimulates the production and function of white blood cells, enhancing both innate and adaptive immune responses. Trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2), which is overexpressed in certain cancers such as HER2-positive breast cancer. The rationale for combining these agents lies in their complementary mechanisms—trastuzumab directly targets HER2-positive tumor cells, while sargramostim may enhance anti-tumor immunity by modulating immune cell activity and serving as an immunotherapy adjuvant. This combination has been explored primarily in clinical trials for HER2-positive breast cancer, particularly to improve immune response or overcome resistance to standard therapies[1][8][10].
