saruparib + abiraterone

**Saruparib + abiraterone** is an investigational oral combination therapy for metastatic prostate cancer, consisting of saruparib (AZD5305), a highly selective poly(ADP-ribose) polymerase (PARP) 1 inhibitor, and abiraterone (abiraterone acetate, Zytiga), an androgen biosynthesis inhibitor. Saruparib targets cancer cells with deficiencies in homologous recombination repair (HRR), especially those with BRCA1 or BRCA2 mutations, by inhibiting PARP1 and impeding DNA repair, thus promoting cancer cell death. Abiraterone reduces androgen production, suppressing the androgen receptor signaling axis that drives prostate cancer progression. The combination is under phase 3 evaluation for metastatic castration-sensitive prostate cancer (mCSPC), including both HRR-mutant and non-HRR-mutant populations.
