Savolitinib + cetuximab + FOLFOX is an investigational combination regimen being evaluated for the first-line treatment of RAS/BRAF wild-type metastatic colorectal cancer (mCRC). The regimen consists of **savolitinib** (Orpathys), a selective small-molecule inhibitor of the MET receptor tyrosine kinase; **cetuximab** (Erbitux), a monoclonal antibody targeting the epidermal growth factor receptor (EGFR); and **FOLFOX**, a standard chemotherapy backbone comprising oxaliplatin, leucovorin (folinic acid), and 5-fluorouracil. The rationale for this triplet combination is to simultaneously inhibit the MET and EGFR pathways, as MET amplification or activation is a recognized mechanism of resistance to anti-EGFR therapies like cetuximab in colorectal cancer. By co-targeting these pathways alongside cytotoxic chemotherapy, the regimen aims to improve objective response rates and survival outcomes in patients whose tumors may otherwise bypass EGFR inhibition through aberrant MET signaling.
