savolitinib + famotidine is a combination of two drugs: savolitinib, a potent, oral small molecule inhibitor that selectively targets the proto-oncogene receptor tyrosine kinase MET (mechanism: MET receptor antagonist), and famotidine, an H2 receptor antagonist (mechanism: Histamine H2 receptor antagonist) commonly used to reduce gastric acid secretion. This combination was evaluated to study potential pharmacokinetic interactions, specifically whether famotidine intake (which increases gastric pH) affects the oral absorption and exposure of savolitinib, whose solubility is pH dependent. Studies showed that co-administration of famotidine with savolitinib reduced exposure to savolitinib, but this effect was not considered clinically significant. The combination has not yet been developed for any approved therapeutic intent, but has been evaluated in Phase 1 pharmacology studies[1][2][3].
