Overview

A combination regimen consisting of: - **Stereotactic Body Radiation Therapy (SBRT):** A highly precise, high-dose form of external beam radiation therapy used to target tumors while minimizing damage to surrounding tissues. SBRT is delivered in a small number of sessions (typically 1-5) and is most often used for small or well-defined tumors in organs such as the lung and liver, or for metastatic lesions[1][3][5][7][9]. - **Fruquintinib:** A highly selective oral small-molecule inhibitor of vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3, developed for the treatment of metastatic colorectal cancer and other solid tumors. Approved for use in patients with metastatic colorectal cancer who have failed prior lines of standard therapy[2][4][6]. - **Cadonilimab:** A bispecific monoclonal antibody targeting the immune checkpoint proteins programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), designed to enhance the immune response against tumors. Used for some advanced solid tumors. This **combination therapy** utilizes local tumor ablation via SBRT, antiangiogenic and antitumor effects via fruquintinib, and immunomodulation via dual immune checkpoint blockade with cadonilimab. The regimen is being investigated in advanced/metastatic solid tumors, likely including metastatic colorectal cancer.
