Overview

SC-006 + ABBV-181 is a combination therapy consisting of **SC-006**, an antibody-drug conjugate (ADC) targeting DNA and **ring finger protein 43 (RNF43)**, and **ABBV-181** (also known as budigalimab), a humanized recombinant IgG1 monoclonal antibody that inhibits **programmed cell death protein 1 (PD-1)**. SC-006 acts as a DNA inhibitor and RNF43 inhibitor, while ABBV-181 (budigalimab) blocks immune checkpoint PD-1 to enhance antitumor immune responses. This combination was developed for treatment of advanced colorectal cancer and was investigated in a Phase 1 clinical trial as both monotherapy and combination therapy. Development in this indication was discontinued after thrombocytopenia limited dose escalation below predicted efficacy[1][2][3][4][5][6].
