Overview
SCB-2019 (CpG 1018/Alum) is a recombinant protein subunit COVID-19 vaccine developed by Clover Biopharmaceuticals. The vaccine features a recombinant SARS-CoV-2 spike (S) protein stabilized in its native-like prefusion trimeric conformation using Clover's proprietary Trimer-Tag technology, based on the Wuhan-Hu-1 strain. To enhance immunogenicity, the vaccine is formulated with two adjuvants: CpG 1018, a Toll-like receptor 9 (TLR9) agonist developed by Dynavax Technologies, and aluminum hydroxide (alum). In the global Phase 2/3 SPECTRA trial, the vaccine demonstrated 67% overall efficacy against COVID-19 of any severity and 100% efficacy against severe disease and hospitalizations. It was administered as a two-dose regimen via intramuscular injection and received emergency use authorization in China in December 2022.
