Overview

scfDNA + polyethylenimine is an experimental DNA nanocarrier designed to mimic the proinflammatory effects of placental cell-free DNA (cfDNA) observed during pregnancy. It consists of a synthetic mimic of fetal cfDNA (scfDNA) complexed with the cationic polymer polyethylenimine (PEI) to facilitate cytosolic delivery. The drug targets the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway in monocytes and macrophages. By activating the cGAS-STING-IRF signaling axis, it induces a proinflammatory phenotype in tumor-associated macrophages (TAMs), potentially overcoming the immunosuppressive tumor microenvironment. This pregnancy-inspired approach is being developed as a strategy for cancer immunotherapy, specifically for reprogramming myeloid cells within the tumor.
