SCH-58500 + paclitaxel + carboplatin is an experimental combination therapy evaluated in clinical trials, particularly for ovarian and non-small cell lung cancer (NSCLC). SCH-58500 is a replication-defective adenoviral vector encoding the wild-type human p53 tumor suppressor gene, intended for gene replacement therapy to restore p53 function in cancer cells. Paclitaxel and carboplatin are standard chemotherapeutic agents routinely used in the treatment of various solid tumors. The rationale for combining these drugs is to enhance tumor cell killing through both restoration of p53-mediated apoptosis (by SCH-58500) and cytotoxic effects (by paclitaxel and carboplatin). Clinical investigations found this combination was feasible and generally safe, but substantial additive clinical benefits over chemotherapy alone were not demonstrated in controlled studies.
