SCIB1 + pembrolizumab is an investigational combination drug regimen used in advanced melanoma, consisting of the DNA cancer vaccine **SCIB1** and the immune checkpoint inhibitor **pembrolizumab**. SCIB1 is a DNA plasmid vaccine that encodes epitopes from TRP-2 and gp100, engineered into an antibody framework to enhance targeting and activation of dendritic cells. This aims to stimulate both CD4 and CD8 T cell anti-tumor immunity. Pembrolizumab is a monoclonal antibody checkpoint inhibitor targeting PD-1, which acts to release suppressed anti-tumor T cell responses. The combination is hypothesized to synergistically enhance tumor immune responses and improve treatment durability over pembrolizumab alone. It is currently under investigation in a Phase 2 trial for patients with unresectable or advanced melanoma[1][3][5][6][7].
