SD-101 + nivolumab** is a combination therapy consisting of two drugs: SD-101, a synthetic CpG oligonucleotide that acts as a Toll-like receptor 9 (TLR9) agonist, and nivolumab, a monoclonal antibody targeting the programmed cell death protein 1 (PD-1) checkpoint on immune cells. SD-101 is administered intratumorally to stimulate local immune activation by inducing type I interferons and promoting dendritic cell activation, resulting in increased T-cell infiltration and tumor immunogenicity. Nivolumab is given systemically to block the PD-1 pathway, thereby enhancing anti-tumor T-cell activity and overcoming immune suppression in the tumor microenvironment. This combination aims to synergistically boost anti-tumor immunity and has been investigated in early clinical trials for metastatic pancreatic cancer and unresectable liver metastases, among other indications.
