SD-101 + nivolumab + relatlimab

SD-101 + nivolumab + relatlimab is a combination immunotherapy regimen under clinical investigation primarily for melanoma. SD-101 is a synthetic oligonucleotide acting as a CpG-C class Toll-like receptor 9 (TLR9) agonist, designed to activate plasmacytoid dendritic cells and initiate both innate and adaptive immune responses, especially leading to interferon production and increased tumor antigen presentation. Nivolumab is a monoclonal antibody that blocks programmed death-1 (PD-1), reinvigorating exhausted T cells to recognize tumor cells. Relatlimab is a monoclonal antibody targeting lymphocyte activation gene-3 (LAG-3), another checkpoint on T cells, further enhancing anti-tumor T cell function. The combined effect is enhanced immune response within the tumor microenvironment, aiming to overcome mechanisms of tumor resistance to PD-1 inhibitors and stimulate robust, durable anti-tumor immunity[2][3][4][5][7].
