SEA-CD70 + azacitidine is an investigational combination therapy for myeloid malignancies, specifically higher-risk myelodysplastic syndromes (HR-MDS) and acute myeloid leukemia (AML). SEA-CD70 (PF-08046040) is a humanized, nonfucosylated, CD70-directed IgG1 monoclonal antibody with enhanced effector function, developed to target CD70, which is expressed on myeloid blasts. Azacitidine is a hypomethylating agent that increases CD70 expression, potentially augmenting the activity of SEA-CD70. Preclinical data and early-phase clinical trials show that the combination is well tolerated, with manageable adverse events and preliminary clinical efficacy in patients with untreated HR-MDS. The combination is being evaluated in multi-part, dose-finding and expansion studies, with plans for Phase III development in HR-MDS.
