This is a combination therapy consisting of three agents: **SEA-CD70**, **azacitidine**, and **venetoclax**. SEA-CD70 is an investigational, humanized, non-fucosylated monoclonal antibody that targets CD70, a protein aberrantly expressed on malignant myeloid blasts, with enhanced effector functions mediated by sugar-engineered antibody (SEA) technology. Its mechanisms include antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), antibody-dependent cellular phagocytosis (ADCP), and blockade of the CD70-CD27 interaction, potentially reducing malignant blast proliferation and immune evasion[1][3][7]. Azacitidine is a hypomethylating agent that inhibits DNA methylation and is commonly used in the treatment of myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML)[2][6]. Venetoclax is a small molecule BCL-2 inhibitor used to induce apoptosis in malignant cells, especially in AML and MDS[4][6]. This triplet regimen is under investigation for the treatment of myeloid malignancies including myelodysplastic syndromes and acute myeloid leukemia, particularly in relapsed or refractory disease settings[1][3][4].
