Augustine Therapeutics is developing a second-generation, peripherally-restricted small molecule inhibitor of Histone Deacetylase 6 (HDAC6) specifically for the treatment of cardiometabolic diseases. This program utilizes a novel non-hydroxamate and non-hydrazide chemotype designed to selectively inhibit the catalytic activity of the cytosolic enzyme HDAC6 while preserving its beneficial non-catalytic functions. This approach aims to avoid the toxicity and poor drug-like properties associated with earlier generations of HDAC6 inhibitors. By targeting HDAC6 in peripheral tissues, the drug candidate is intended to modulate cellular processes such as microtubule stability and transport without crossing the blood-brain barrier. The program is currently in the advanced lead optimization phase of preclinical development.
