Secukinumab + SG3249 is an isotype control antibody-drug conjugate (ADC) primarily utilized in preclinical oncology research. It is composed of the humanized monoclonal antibody **secukinumab**, which targets the soluble cytokine **interleukin-17A (IL-17A)**, site-specifically conjugated to the **pyrrolobenzodiazepine (PBD)** dimer toxin-linker **SG3249** (also known as **tesirine**). Because its target, IL-17A, is a secreted protein and not expressed on the surface of tumor cells, this conjugate does not bind to or undergo internalization in cancer cells. It serves as a critical negative control in studies evaluating the efficacy and safety of other PBD-based ADCs (such as **TR1801-ADC** or **PCA062**), allowing researchers to distinguish between target-mediated antitumor activity and non-specific toxicity associated with the PBD payload in patient-derived xenograft (PDX) models and cell line assays.
