Overview
Segesterone acetate + testosterone is a transdermal gel formulation under development by the Population Council and the National Institute of Child Health and Human Development (NICHD) as a male hormonal contraceptive. The combination consists of segesterone acetate (Nestorone), a potent progestin that acts as a progesterone receptor agonist to suppress the hypothalamic-pituitary-gonadal axis, and testosterone to maintain physiological androgen levels. By inhibiting the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), the gel effectively suppresses spermatogenesis. The daily, self-administered topical application is designed to provide a reversible and effective contraceptive method for men with fewer side effects than injectable alternatives.
