SEL-302 is an investigational combination therapy developed by Selecta Biosciences for the treatment of the Mut subtype of methylmalonic acidemia (MMA). It consists of two distinct components: **MMA-101**, an adeno-associated virus serotype 8 (AAV8) gene therapy vector, and **ImmTOR** (also known as SEL-110), a proprietary immunomodulatory agent. MMA-101 is designed to deliver a functional copy of the methylmalonyl-CoA mutase (MMUT) gene to the liver to address the underlying enzyme deficiency. ImmTOR consists of rapamycin-containing nanoparticles and is administered prior to the gene therapy to induce immune tolerance to the AAV vector. This approach aims to mitigate unwanted immune responses, such as the formation of anti-AAV antibodies, which typically prevent the redosing of gene therapies. The therapy is currently being evaluated in Phase 1/2 clinical trials for pediatric patients with isolated MMA caused by MMUT deficiency.
