This is a combination therapy consisting of three components: 1. **Selinexor**: A selective XPO1 inhibitor that blocks the export of tumor suppressor proteins and growth regulatory proteins from the nucleus to cytosol, restoring tumor-suppressive pathways. It has been approved as part of regimens to treat multiple myeloma and diffuse large B-cell lymphoma. 2. **Anti-PD-1 antibody**: Immune checkpoint inhibitors that block the PD-1 pathway, enhancing T-cell immune responses against cancer cells. Examples include tislelizumab, pembrolizumab, and nivolumab. 3. **P-GemOx**: A chemotherapy regimen consisting of pegaspargase, gemcitabine, and oxaliplatin, commonly used in the treatment of extranodal natural killer/T-cell lymphoma (ENKTL). Preclinical studies have shown that selinexor can sensitize cancer cells to anti-PD-1 antibodies, potentially enhancing their efficacy. The combination of selinexor with anti-PD-1 antibodies has shown promising results in patients with natural killer/T-cell lymphoma who had exhausted other treatment options.
