Overview

Selinexor + carmustine is an experimental combination therapy comprising selinexor, a selective inhibitor of nuclear export (SINE) that targets exportin-1 (XPO1), and carmustine, an alkylating nitrosourea chemotherapeutic. Selinexor functions as an oral small molecule that blocks the export of tumor suppressor proteins from the nucleus, thereby enhancing apoptosis in cancer cells. Carmustine introduces DNA damage via alkylation, leading to cell death, particularly in rapidly dividing malignant cells. This combination has been investigated in clinical trials for recurrent glioblastoma (rGBM), based on the rationale that dual inhibition of nuclear export and DNA integrity may provide synergistic anti-tumor efficacy.[3][4][5]
