rejuvenates tumor suppressor proteins and induces apoptosis. - *Dexamethasone* is a synthetic glucocorticoid corticosteroid, used primarily for its anti-inflammatory and immunosuppressive effects, and as a backbone in myeloma regimens. - *Belantamab mafodotin* is a first-in-class antibody-drug conjugate that targets B-cell maturation antigen (BCMA) on myeloma cells. The afucosylated anti-BCMA humanized IgG1 antibody is conjugated to the cytotoxic payload monomethyl auristatin F (MMAF); upon binding and internalization, MMAF disrupts microtubule networks, induces cell cycle arrest at G2/M, and causes apoptosis. Belantamab mafodotin further promotes immunogenic cell death through antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis[2][4][8]. This combination is being studied as a potential regimen for patients with RRMM, particularly those who have failed multiple prior lines of therapy[1][7].
