Overview

This is a combination regimen consisting of three oral small molecule drugs—selinexor, ixazomib, and lenalidomide—used in the treatment of multiple myeloma. - Selinexor is a first-in-class selective inhibitor of nuclear export (SINE) that blocks exportin 1 (XPO1), leading to accumulation of tumor suppressor proteins in the nucleus and promoting apoptosis in cancer cells. - Ixazomib is an oral proteasome inhibitor that disrupts protein degradation pathways essential for cancer cell survival. - Lenalidomide is an immunomodulatory agent with antiangiogenic and direct antineoplastic effects, modulating the immune response against myeloma cells. This combination leverages distinct mechanisms to target multiple pathways involved in myeloma cell growth and survival, aiming for synergistic antitumor activity[2][4][5][8].
