Overview

**Selinexor + tislelizumab** is an investigational combination drug regimen consisting of selinexor, an oral small molecule inhibitor of exportin 1 (XPO1), and tislelizumab, a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1)[1][3][5].\nSelinexor selectively inhibits nuclear export by binding to XPO1, resulting in accumulation of tumor suppressor proteins in the nucleus and promoting cancer cell apoptosis[1][5].\nTislelizumab blocks the PD-1 pathway, enhancing anti-tumor immune response by preventing inhibition of T-cell activity[1][3][5].\nThis combination is being evaluated primarily for its synergistic effect in relapsed/refractory T-cell and natural killer (NK)-cell lymphomas, including cases with central nervous system involvement[1][2][3][5]. Early clinical data suggest deep and durable anti-tumor responses may be achievable with manageable toxicity[1][3].
