Drug intelligence / Profile preview
**Semaglutide + ALK7 siRNA** is an investigational combination regimen being evaluated for the treatment of obesity and metabolic disorders. The combination pairs semaglutide, a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist developed by Novo Nordisk, with an activin receptor-like kinase 7 (ALK7) small interfering RNA (siRNA) therapeutic (such as Arrowhead's ARO-ALK7, SanegeneBio's SGB-ALK7, or Alnylam's ALN-2232). Semaglutide primarily acts on the central nervous system to suppress appetite and reduce food intake, while the ALK7 siRNA selectively silences the expression of the *ACVR1C* gene in adipose tissue. ALK7 acts as a metabolic brake that suppresses lipolysis and promotes fat storage; silencing ALK7 enhances lipolysis and energy expenditure. Preclinical studies have demonstrated that combining ALK7 siRNA with semaglutide produces additive weight loss and fat mass reduction while significantly preserving lean muscle mass, addressing a key limitation of GLP-1 monotherapy.
