Drug intelligence / Profile preview
Semaglutide + TAU refers to the adjunctive use of the glucagon-like peptide-1 (GLP-1) receptor agonist semaglutide alongside a patient's existing antipsychotic medication regimen, referred to in clinical trials as 'Treatment As Usual' (TAU). This specific combination is being evaluated in the ESCaMS trial (Efficacy and Safety of Semaglutide Versus Placebo on Cardiometabolic Profile in Patients With Schizophrenia With Metabolic Syndrome), led by the All India Institute of Medical Sciences, Bhubaneswar. The intervention involves adding oral semaglutide (titrated to 3 mg once daily) to ongoing second-generation antipsychotic therapy to improve the cardiometabolic profile, reduce 10-year cardiovascular risk (QRISK3), and manage weight gain and insulin resistance associated with antipsychotic use. Semaglutide works by mimicking the GLP-1 hormone to regulate blood sugar and appetite, while the antipsychotic component typically manages psychiatric symptoms through dopamine and serotonin receptor antagonism.
