sEphB4-HSA + nab-paclitaxel is an investigational combination therapy consisting of a soluble EphB4-human serum albumin (HSA) fusion protein and an albumin-bound nanoparticle formulation of paclitaxel. Developed by the University of Southern California, sEphB4-HSA acts as a decoy receptor that binds to the ligand EphrinB2, thereby inhibiting bidirectional signaling through the EphB4 receptor. This pathway is a key regulator of tumor angiogenesis, vascular remodeling, and tumor cell proliferation. Nab-paclitaxel (Abraxane) is a cytotoxic microtubule stabilizer that inhibits cell division. The combination is being evaluated in a Phase Ib clinical trial for various advanced solid tumors, including pancreatic adenocarcinoma, head and neck squamous cell carcinoma, non-small cell lung cancer, and biliary tract cancers (cholangiocarcinoma and gallbladder carcinoma), often in further combination with other chemotherapeutic agents like gemcitabine.
