This is a combination regimen consisting of three anticancer agents: **seribantumab**, a fully human monoclonal antibody that targets human epidermal growth factor receptor 3 (HER3, also known as ERBB3); **cetuximab**, a monoclonal antibody targeting the epidermal growth factor receptor (EGFR); and **irinotecan**, a small molecule topoisomerase I inhibitor. - **Seribantumab** binds HER3/ERBB3, blocking neuregulin 1 (NRG1/HRG)-mediated activation and downstream signaling, including PI3K/AKT and MAPK pathways, which are associated with cancer cell proliferation, survival, and drug resistance[1][3][5][7]. - **Cetuximab** blocks EGFR signaling, directly inhibiting tumor growth[2][4][6]. - **Irinotecan** inhibits topoisomerase I, leading to DNA damage and apoptosis in dividing cells[2][4][6]. The rationale for combining these agents is to simultaneously inhibit multiple, frequently compensatory, growth and survival pathways—potentially improving efficacy in solid tumors, including colorectal cancer and other malignancies where HER3, EGFR, and their ligands are active.
