Hetronifly + pemetrexed + carboplatin is a combination chemoimmunotherapy regimen approved in the European Union for the first-line treatment of adult patients with unresectable locally advanced or metastatic non-squamous non-small cell lung cancer (NSCLC) lacking EGFR, ALK, or ROS1 alterations. The regimen combines serplulimab (Hetronifly), a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, with two standard chemotherapeutic agents: pemetrexed, a folate analog metabolic inhibitor, and carboplatin, a platinum-based DNA alkylating agent. Developed by Shanghai Henlius Biotech and commercialized in Europe by Accord Healthcare, this combination works synergistically by utilizing chemotherapy to induce tumor cell death and antigen release, while serplulimab blocks the PD-1/PD-L1 pathway to restore and enhance T-cell-mediated anti-tumor immune responses.
