Serplulimab + SOX chemotherapy is a combination regimen consisting of the anti-PD-1 monoclonal antibody serplulimab and the SOX chemotherapy doublet (S-1 and oxaliplatin). Serplulimab, developed by Shanghai Henlius Biotech, is a humanized IgG4 monoclonal antibody that blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby revitalizing the anti-tumor immune response. SOX chemotherapy comprises S-1 (an oral fluoropyrimidine consisting of tegafur, gimeracil, and oteracil) and oxaliplatin (a platinum-based cytotoxic agent). This combination is primarily utilized as a neoadjuvant or perioperative treatment for locally advanced, resectable gastric and gastroesophageal junction (GEJ) adenocarcinomas. Clinical evidence, including the Phase 3 ASTRUM-006 trial, suggests that adding serplulimab to SOX chemotherapy significantly improves event-free survival and pathological response rates compared to chemotherapy alone.
