Overview
Sertraline + aripiprazole is a combination therapy being evaluated for the treatment of obsessive-compulsive disorder (OCD). This specific treatment strategy involves the augmentation of the selective serotonin reuptake inhibitor (SSRI) sertraline with the atypical antipsychotic aripiprazole. It is a key intervention arm in the PROCEED (Pharmacotherapy Options Following Unsatisfactory Initial Treatment in OCD) trial, a phase 2 multi-center study led by the Shanghai Mental Health Center. The combination is intended for patients who do not achieve remission after an initial 12-week course of sertraline monotherapy. Sertraline works by inhibiting the serotonin transporter, while aripiprazole acts as a partial agonist at dopamine D2 and serotonin 5-HT1A receptors and as an antagonist at 5-HT2A receptors. This dual mechanism is hypothesized to enhance therapeutic response in treatment-resistant OCD cases.
