Drug intelligence / Profile preview
This is a combination of seven antidepressant drugs: **sertraline**, **escitalopram**, **fluoxetine**, **duloxetine**, **paroxetine**, **venlafaxine**, and **fluvoxamine**. All seven are centrally acting antidepressants classified as either selective serotonin reuptake inhibitors (SSRIs; sertraline, escitalopram, fluoxetine, paroxetine, fluvoxamine) or serotonin-norepinephrine reuptake inhibitors (SNRIs; venlafaxine, duloxetine). These drugs exert their antidepressant effects primarily by inhibiting the reuptake of serotonin (5-HT) in the central nervous system, increasing synaptic serotonin levels. SNRIs also inhibit norepinephrine reuptake. The combination of multiple agents from similar classes is generally not standard practice due to increased risk of adverse effects, most notably serotonin syndrome, a potentially life-threatening condition. This combination would be considered in clinical practice only in extremely rare, resistant, or experimental situations, and generally is not recommended due to safety risks[1][5][6][9]. Mechanism of action for all included drugs involves inhibition of serotonin transporter (SERT), with duloxetine and venlafaxine also possessing significant norepinephrine transporter (NET) inhibition. Primary indications for each agent individually include major depressive disorder, generalized anxiety disorder, obsessive-compulsive disorder, panic disorder, and other mood or anxiety-related conditions[1][5][3].
